Effects on Ability to Drive and Use Machines: There is no evidence that CREON has any effect on the ability to drive or operate machines.

Pregnancy
There are no adequate data from the use of CREON MINIMICROSPHERES in pregnant women. Animal studies are insufficient with respect to effects on pregnancy and embryonal/foetal development, parturition and postnatal development. The potential risk for humans is unknown. CREON MINIMICROSPHERES should not be used during pregnancy or lactation unless clearly necessary.

CREON 10 MINIMICROSPHERES
Each opaque, brown/colorless-transparent, hard gelatin capsule, imprinted in white with SOLVAY and 1210, contains: buff-colored enteric-coated minimicrospheres of pancreatin corresponding to 10 000 USP units of lipase, 33 200 USP units of amylase and 37 500 USP units of proteases. Nonmedicinal ingredients: dibutylphthalate, dimethicone 1000, gelatin, hydroxypropyl methylcellulose, phthalate, black iron oxide, red iron oxide, yellow iron oxide, light mineral oil, polyethylene glycol 4000 and titanium dioxide. Each opaque, orange/colorless, transparent hard gelatin capsule, imprinted in white with SOLVAY and 1220, contains: buff-colored enteric-coated minimicrospheres of pancreatin corresponding to: 20 000 USP units of lipase, 66 400 USP units of amylase and 75 000 USP units of proteases. Nonmedicinal ingredients: dibutylphthalate, dimethicone 1000, gelatin, hydroxypropyl methylcellulose phthalate, red iron oxide, yellow iron oxide, light mineral oil, polyethylene glycol 4000 and titanium dioxide. CREON 25 MINIMICROSPHERES
Each opaque, orange/colorless-transparent hard gelatin capsule, contains: buff-colored enteric-coated minimicrospheres of pancreatin corresponding to: 25 000 USP units of lipase, 74 000 USP units of amylase and 62 500 USP units of proteases. Nonmedicinal ingredients: dibutylphthalate, dimethicone 1000, gelatin, red iron oxide, yellow iron oxide, macrogol 4000, methylhydroxypropylcellulose phthalate, paraffinum perliquidum and titanium dioxide. Gluten-, lactose- and tartrazine-free. Bottles of 100.
Store in tightly closed, light resistant container at controlled room temperature (15 to 30°C). Protect from moisture. Do not refrigerate. Keep out of reach of children.
CREON 5 MINIMICROSPHERES
Each opaque, blue/orange hard gelatin capsule, imprinted in white with SOLVAY and 1205, contains: buff-colored enteric-coated minimicrospheres of pancreatin corresponding to: 5000 USP units of lipase, 16 600 USP units of amylase and 18 750 USP units proteases. Nonmedicinal ingredients: dibutylphthalate, dimethicone 1000, FD&C Blue #2, gelatin, hydroxypropyl methylcellulose phthalate, red iron oxide, yellow iron oxide, light mineral oil, polyethylene glycol 4000 and titanium dioxide. Gluten-, lactose- and tartrazine-free. Bottles of 100.
Warnings
Rarely, cases of hyperuricosuria and hyperuricaemia have been reported with very high doses of pancreatin.
Perianal irritation, and rarely, inflammation, could occur when large doses are used.
Contraindications
Patients with known hypersensitivity to porcine proteins, pancreatic enzymes, or to any excipients. Do not use during the early stages of acute pancreatitis.
Adverse Effects
Skin and Subcutaneous Tissue Disorders
Allergic or hypersensitivity reactions of the skin were reported (uncommon).
Gastrointestinal Disorders
abdominal discomfort/pain (common), constipation (uncommon), abnormal stool (uncommon), diarrhea (uncommon), and nausea/vomiting (uncommon) have been reported, but are also often symptoms of the diseases treated. With high doses perianal irritation/inflammation have also been reported.
Strictures of the ileo-caecum and large bowel (fibrosing colonopathy) and colitis have been reported in patients with cystic fibrosis taking high doses of pancreatin preparations. Case control studies did not reveal evidence of an association between Creon and the appearance of fibrosing colonopathy. As a precaution, unusual abdominal symptoms or changes in abdominal symptoms should be reviewed to exclude the possibility of colonic damage, especially if the patient is taking in excess of 10 000 units of lipase/kg/day.
Metabolism and Nutritional
With high doses, cases of hyperuricosuria and hyperuricaemia have been reported.
Indications
For replacement therapy where digestion is not adequate due to pancreatic exocrine insufficiency as in cystic fibrosis, chronic pancreatitis, steatorrhoea, post pancreatectomy, post gastrointestinal bypass surgery (e.g., gastroenterostomy) and ductal obstruction from neoplasm.
Dosage
CREON 5 Minimicrospheres: Initially, two to four capsules with each meal and two to four capsules with each snack.
CREON 10 MINIMICROSPHERES: Initially one or two capsules with each meal and one capsule with each snack.
CREON 20 MINIMICROSPHERES: Initially, one capsule with each meal and one capsule with each snack.
CREON 25 MINIMICROSPHERES: Initially, one capsule with each meal and one capsule with each snack.
Dosage increases, if required, should be added slowly, with careful monitoring of response and symptomatology.
It is important to ensure adequate hydration of patients at all times during dosing with pancreatic enzymes.
The capsules should be swallowed whole. Where swallowing the capsules is difficult, they may be opened and the minimicrospheres taken with soft food or fluid. Any mixture of the minimicrospheres with food or liquids should be used immediately and not stored; otherwise dissolution of the enteric coating may occur.
To protect the enteric coating, the minimicrospheres must not be crushed or chewed.
The product should not be administered after the expiry date printed on the package.
Overdose
Symptoms
Extremely high doses of pancreatin have been reported to be associated with hyperuricosuria and hyperuricaemia.
Treatment
Most cases of overdose respond to supportive measures, including stopping enzyme therapy and ensuring adequate rehydration.
